My real answer is Star Trek 2: The Wrath of Khan, with Kirstie Alley as the Cheers cast member and Spock as the alien (or Khan's Pets if you want to go the gross route).

Star Wars: Episode IV - A New Hope, with Han as the agnostic ("Hokey religions and ancient weapons are no match for a good blaster at your side, kid."), along with plenty of alien characters (e.g. Jawas, Tusken Raiders, Greedo and other Mos Eisley patrons, and of course, Chewie).
